The zona reaction of hamster and mouse eggs: production in vitro by a trypsin-like protease from cortical granules.

چکیده
The cortical granules of hamster ova were ruptured in vitro by electrical stimulation (standard treatment was 150 V, 1\m=.\0msec). Approximately 8 min were required for the cortical granule material so released to induce a complete zona reaction. The active material from the cortical granules was released into the medium from vitelli and was shown to act directly on the zona pellucida and on mouse, as well as

Heat sensitivity of the cortical granule protease from hamster eggs.
Previous studies (Gwatkin, Williams, Hartmann & Kniazuk, 1973) showed that the zona reaction of the hamster ovum is produced by a cortical granule protease, which inactivates the species-specific receptor-for-spermatozoa located in the zona pellucida. This receptor was earlier shown to be sensitive to low concentrations of pancreatic trypsin (Hartmann & Gwatkin, 1971). Several years ago Austin ...
متن کاملI-13 Infertility with Impaired Zona Pellucida Adhesion of Spermatozoa from Mice LackingTauCstF-64
Background: Fertilization is a multistep process requiring spermatozoa with unique cellular structures and numerous germ cell-specific molecules that function in the various steps. In the highly coordinated process of male germ cell development, RNA splicing and polyadenylation help regulate gene expression to ensure formation of functional spermatozoa. Male germ cells express tauCstF-64 (Cstf2...

Incubation of hamster eggs with trypsin or chymotrypsin, but not with a series of glycosidases or lipases, prevents capacitated spermatozoa from binding to the zona pellucida so that fertilization is prevented (Hartmann & Gwatkin, 1971 ; Gwatkin, Williams & Andersen, 1973). Inactivation of the binding sites for spermatozoa on the zona pellucida by a trypsin-like protease released by discharge o...
متن کاملIntactness of zona pellucida does not affect the secretion of a trypsin-like protease from mouse blastocyst.
Assisted hatching (AH), which is known to improve the hatching potential of mammalian embryos, has been used to increase the pregnancy rate in in vitro fertilization cycles. However, the effect of AH on a trypsin-like protease, which is known to be associated with the hatching process, has not been studied. In this study, we evaluate whether the intactness of zona pellucida affects the secretio...
متن کاملO-8: Molecular Mechanisms of Membrane Fusion Involved in Fertilization
Background: Assisted fertilization procedures are a currently widespread practice to regulate reproduction in humans and animals. The arising question is why the human being manipulating gametes to generate new individuals, if we do not understand yet the molecular mechanism of fertilization?. Successful completion of fertilization in mammals is dependent on three membrane fusion events: 1. the...
